Extracellular crosslinking mass spectrometry reveals HLA class I – HLA class II interactions on the cell surface

چکیده
Human Leukocyte Antigen (HLA) complexes are critical cell-surface protein assemblies that facilitate T-cell surveillance of almost all cell types in the body. While receptor binding to HLA class I and II is well-described with detailed structural information, nature cis interactions within plasma membrane surveyed cells remains be better characterized, as protein-protein environment technically challenging profile. Here we performed extracellular chemical crosslinking on intact antigen presenting specifically elucidate present external membrane. We found crosslink dataset was dominated by inter- intra-protein crosslinks involving molecules, which enabled not only construction an HLA-centric interaction map, but also revealed multiple modes – further modeling based crosslinker distance restraints. Collectively, our data demonstrate molecules colocalize can densely packed

HLA Class I and Class II Genes Distribution of the Sistanis in Iran
Background: The high polymorphism in the human leukocyte antigen (HLA) genes can be used as an identity of individuals to compare with other populations. This extreme polymorphism in the HLA system is accountable for the differences in alleles and haplotypes among ethnic groups, populations, and the inhabitants of many regions. Objective: To define the frequency of HLA alleles and haplotypes am...
متن کاملAssociation between HLA-Class I and HLA-Class II Alleles and Mycobacterium Tuberculosis Infection in Iraqi Patients from Baghdad City
Background: Pulmonary tuberculosis (PT) is one of the endemic diseases in Iraq, and among the suggested predisposing factors are alleles of the human leukocyte antigen (HLA) system. We sought to investigate the association between HLA-class I (A and B) and -class II (DR and DQ) alleles in a sample of PT Iraqi patients. Methods: lymphocytes of 105 PT patients and 40 controls were phenotyped for H...
متن کاملCell surface expression and function of an HLA class II molecule with class I domain configuration
Recombinant major histocompatibility complex (MHC) class II molecules were expressed with extracellular polypeptide domains reorganized to form heavy (H) and light (L) chains (alpha 1-beta 1-beta 2 and alpha 2) analogous to class I. Accurate protein folding and dimerization is demonstrated by the ability of this 3+1-DR1 construct to bind class II-restricted peptides and stimulate CD4+ T cells. ...
متن کاملULCERATIVE COLITIS AND HLA CLASS II PHENOTYPING IN IRAN
Ulcerative colitis (UC) is a nonspecific acute and chronic inflammatory bowel disease that diffusely involves the colonic mucosa. The etiology of UC has not yet been elucidated fully. However, many studies have found that immunologic disorders may play a role in the pathogenesis of UC. In addition, due to an increased frequency of UC in families, especially an increased monozygotic compared...
متن کاملHLA CLASS I IN IRANIAN P SORIATIC PATIENTS
The association of HLA and disease varies in different populations. We have . studied the association of HLA class I in Iranian psoriatic patients, in order to compare this association with other reports. Fifty-one Iranian patients with psoriasis were HLA typed. The frequency of HLA antigens in patients and patient subgroups, based on clinical patterns, age of onset, family history and prov...
